The Princess Máxima Center for Pediatric Oncology is a new research hospital, which concentrates healthcare, research and education with regard to cancer in children in a single location in Utrecht. The institute aims to provide the highest level of care for all children with cancer and has the ambition to cure all children of cancer and to significantly reduce unwanted side effects. The center brings together the best possible care and scientific research, creating a unique interdisciplinary institute for pediatric oncology in Europe.
The research group of Jan Molenaar at the Princess Maxima Center is specialized in translational research in pediatric solid tumors with a focus on Neuroblastoma. To this purpose we have developed an experimental pipeline which starts with basic molecular biologic research and ends with implementation of new treatment options for pediatric cancer patients. Crucial in this pipeline is the use of high throughput analysis. We are running the compound screening facility in the Maxima and we have initiated the iTHER precision medicine program for pediatric cancer, which has just included 100 patients. Recently we have started a research line on immunologic model systems and interventions in Neuroblastoma. We have been typing the tumor micro environment (TME) using scRNAseq, and immune profiling. We have generated neuroblastoma co-culture systems (organoids with immune cells) and we have initiated Tumor Infiltrating Lymphocytes testing to prepare for potential therapeutic interventions.
This research project aims at in-dept typing of the tumor micro environment and development of immune interventions based on these analyses. We have access to tumor materials from various stages in the disease course (diagnosis, treatment and relapse). Samples from different stages will be used to perform scRNAseq analysis, immune profiling, proteomics of the MHC bound ligandome, TCR sequencing and we will do the standard NGS on DNA and RNA. As such we will get an in depth overview of the immune environment at various stages of tumor development. In addition, we will generate model systems from all stages including organoids for co-culture, and preferably humanized mice. The high-throughput analysis will be used to identify 3 types of interventions 1) Potential active inhibitory checkpoints, 2) Neo-epitopes and corresponding active TILs, and 3) potential tumor specific epitopes for antibody mediated interventions. Most analysis can be performed in our institute. In addition, we have close collaborations in the immunology field with the group from Stefan Nierkens from the UMCU/PMC, Utrecht, the group from Jurgen Kuball at the UMC Utrecht, and several other research groups.
Education and skills
For the offered position, we are looking for a highly enthusiastic and motivated technician with expertise in cancer immunology research techniques. Training level, at least HLO. You have knowledge on cell culture, the use of immune cells in lab experiments, FACs analysis/sorting of immune cells and other lab techniques for immunology research. You like to collaborate and support other researchers and you are keen on clear communication with colleagues about research methods and results.
We offer a full time position (36 hours) position in an inspiring and multidisciplinary research environment. You will initially be appointed for a period of 1 year, with a possible extension to 2 or more years (full-time). Your gross monthly salary will depend on experience and background, starting in scale 45 with 8,33% gross monthly salary holiday allowance and 8,33 % end-of-year bonus. The Princess Máxima Center operates according to the collective labor agreement ‘cao algemene ziekenhuizen’.
