Tytgat group
The focus of the Tytgat group is to improve cure and quality of life in paediatric solid tumors, with emphasis on neuroblastoma patients. Neuroblastoma is the most common extracranial solid tumour in children and it is derived from precursor cells of the sympathetic nervous system. The prognosis for children with neuroblastoma is highly variable, ranging from spontaneous remission to a 5-year survival rate of only 50%. The discovery that many tumours become dependent on specific metabolic processes has increased interest in developing therapeutic strategies that target cancer metabolism. With respect to neuroblastoma, little is currently known regarding the role of neuroblastoma-specific genetic features such as MYCN gene amplification, telomere status, and copy number alterations on the tumour’s metabolic profile. In this project, we would like to identify a neuroblastoma-specific metabolic profile and key metabolic pathways using a metabolomics approach. Potential therapeutic targets will be validated using in vitro studies using cell lines and patient-derived organoids and cell lines with Dox-inducible expression of genes involved in major oncogenic and differentiation pathways.

Within the Tytgat group, urinary excretion of catecholamines is being studied by our team, within our own cohort and in international collaboration. The studies on the metabolic profile and key metabolic pathways will be performed in close collaboration with the catecholamine team. In these projects, we have an intensive collaboration with dr A. van Kuilenburg, Principal Investigator Amsterdam UMC, Laboratory Genetic Metabolic Diseases Amsterdam UMC, and prof N. Verhoeven-duif, head section Metabolic Diagnostics, UMC Utrecht, prof dr J. Molenaar Princess Máxima Center for Paediatric Oncology, Utrecht.

For this vacancy we are looking for an enthusiastic PhD student with a bachelor’s degree in Medical Biology, Life Sciences, Biochemistry or in a related field. The ideal candidate is looking forward to work in an ambitious team. Work experience in a biomedical or analytical laboratory, using molecular biology and mass spectrometry techniques, is essential. This vacancy requires command of both oral and written English. Excellent communication and organization skills as well as the ability to work within a dynamic group are essential.

What we offer
A temporary, full-time PhD position for a total duration of 4 years (36 hours / week) in an ambitious and internationally recognised team. You will initially be contracted for a period of 1 year, after which your performance will be evaluated and the contract may be extended for three more years. You will be working in a very stimulating and cross-disciplinary research environment, combining cutting-edge imaging research in a state-of-the-art clinical trial environment. Your gross monthly salary will depend on experience and background, starting in scale FWG 45/6 with 8,33% gross monthly salary holiday allowance and 8,33 % end-of-year bonus. The Princess Máxima Center operates according to the collective labour agreement ‘cao algemene ziekenhuizen’.
